平板計(jì)數(shù)瓊脂（PCA）培養(yǎng)基的配方[ 青島日水 ]

用途：用于細(xì)菌總數(shù)測(cè)定（GB 4789.2-2010，SN0168-92 ，GB13093-91，SN/T 2098-2008和ISO4833-2003）。

原理：蛋白胨和酵母浸粉作為基礎(chǔ)營(yíng)養(yǎng)物質(zhì)提供一般細(xì)菌生長(zhǎng)所需的氮源、碳源、B族維生素和其它營(yíng)養(yǎng)物質(zhì)；葡萄糖提供生長(zhǎng)代謝所需的能量物質(zhì)；瓊脂作為凝固劑。
 
配制方法:
稱(chēng)取本品23.5g于1L蒸餾水或去離子水中（可按比例增加或減少配制量）,加熱煮沸至完全溶解,分裝,121℃高壓滅菌15分鐘,滅菌結(jié)束后請(qǐng)搖勻,以防瓊脂沉積于器皿底部而凝固,備用。

Tryptone（胰蛋白胨）：5.0g
Yeast Extract（酵母浸粉）：2.5g
Dextrose（葡萄糖）：1.0g
Agar（瓊脂）：15.0g          
pH7.0±0.2(25℃)

Formulation of Plate Count Agar (PCA) medium
Usage: Used for total bacterial count determination (GB 4789.2-2010, SN0168-92, GB13093-91, SN/T 2098-2008 and ISO4833-2003).
Principle: peptone and yeast extract as basic nutrients to provide nitrogen, carbon, B vitamins and other nutrients for general bacterial growth; glucose to provide the energy needed for growth and metabolism; agar as a coagulant.
Method of preparation:
Weigh this product 23.5g in 1L distilled water or deionized water (can be proportionally increased or reduced formulation), heat and boil until completely dissolved, sub-packed, high-pressure sterilization at 121 C for 15 minutes, shake well after sterilization, in order to prevent agar deposition in the bottom of the vessel and solidify, reserve.
